You'll need an Intel Mac and Leopard for this.
Download the iPhone SDK from Apple. You'll need a free ADC account to do it.
Install the developer tools. You can trim down the installer, but you have to include the part called iPhone SDK.
Go to this folder on your hard drive: /Developer/Platforms/iPhoneSimulator.platform/Developer/Applications/ and find the application called iPhone Simulator.
Put it in your dock and enjoy. It uses your network connection to access the internet so it's extra speedy and you can even get some great screenshots now. -

A good resource, though you have to jump through a lot of hoops. I'm betting someone knows an easier way.
I've actually been thinking about a template designed specifically for the iPhone. Digg and Facebook's interfaces are just so slick. And they scale incredibly well, I gotta think they'd be useful for a lot of sites. -

Edit: I think there's probably a lot of resources out there for this. I found iPhone Tester pretty quickly. -
